Abstract

The genetic progress obtained with trangenesis required the development and consolidation of reproductive techniques such as in vitro embryo production and gamete cryopreservation. With the diversity of new models and genetic lines, it became critical to optimize the use of experimental techniques of cryopreservation of semen in different animal species. This report had as goal to evaluate the efficacy of two different collect techniques about semen quality index: ejaculated volume, motility,vigor and spermatic cells concentration of SWISS-ALBINO mouse (heterogenetic). The technique 1 (T1) used the anatomical structures rupturement, from 5 to 7 cuts, helped by na hipodermic needle and the technique 2 (T2) was done by the epididymis tail pression, directed to the  ductus deferns, helped by two anatomical tweezers, in order to get the suspension. It was realized that the motility and the spermatic concentration in T1 (74,0% and 178,4x106) was superior (P<0,01) to T2 (63,5% e 105,4x106) in this way. The vigor variable showed difference (P<0,05) among the treatments. The semen collect technique through cuts is more advantageous then the structures pression technique, because the results brought less negative effects by the semen quality índex and with a raised collected spermatozoa number.
